do you have a wireless card installed? A problem ive heard before is that if the wireless card is enabled then it polls for new networks every 60 seconds automatically, which can cause lag spikes. Try disabling it!

If thats not it try systematically going through your running processes list and killing what youre certain you dont need.

even if your not certain then kill it anyways, if the pc dies then dont kill it when you reboot.

Yes but while youre connected it searches for other networks in the background.

Expecially in Vista wireless should always be disabled while you play.
